What is NITHYA S-Line Smooth?

NITHYA S-Line Smooth is a specialist preparation for needle mesotherapy produced by Euroresearch Srl (Milan, Italy) — exclusively distributed by PureMED in Poland. It combines three complementary active ingredients: Argireline® (Hexapeptide-8) — a peptide that relaxes facial muscles, type I atelocollagen — a collagen biostimulator, and an amino acid complex — the building blocks of new dermal matrix.

Unlike other NITHYA preparations, S-Line Smooth addresses a specific and often overlooked mechanism of ageing: the dynamic formation of expression wrinkles through repeated facial muscle contractions. It is the only product in the NITHYA line combining myorelaxant action (Argireline®) with simultaneous collagen biostimulation (atelocollagen).

Argireline® (Hexapeptide-8) – mechanism of action at the SNARE level

Argireline® is a trademarked hexapeptide (6 amino acid fragment) with the INCI name Acetyl Hexapeptide-8. It is a synthetic analogue of the N-terminus of the SNAP-25 protein — one of the key proteins of the SNARE complex responsible for releasing acetylcholine at the neuromuscular junction.

The mechanism of action is based on competitive inhibition of the SNARE complex:

• Normally, when a nerve impulse reaches the neuromuscular junction, SNARE complex proteins (SNAP-25, syntaxin, VAMP) bind together, causing fusion of synaptic vesicles with the cell membrane and release of acetylcholine — leading to muscle contraction.
• Argireline® competes with SNAP-25 for its position in the SNARE complex — its presence disrupts the correct formation of the complex, reducing the amount of acetylcholine released.
• Effect: partial, reversible reduction in the contraction strength of facial muscles — muscles still work, but with less intensity. Expression wrinkles become shallower and the formation of new ones is slowed.

This mechanism is fundamentally different from botulinum toxin — BoNT type A permanently cleaves SNAP-25, causing complete blockade of acetylcholine release (the "frozen face" effect). Argireline® works reversibly, without paralysis — the effect is subtler, more natural and gradually fades.

Why is injectable Argireline® more effective than topical cosmetics?

• Topical Argireline® — must penetrate the stratum corneum (lipid barrier), which significantly limits the bioavailability of the peptide molecule in the dermis. The effect is mild and requires months of daily use.
• Argireline® in needle mesotherapy (S-Line Smooth) — delivered directly into the dermis, in the immediate vicinity of neuromuscular junctions. Bioavailability is incomparably higher; the effect is distinct and visible after just 1–2 sessions.

Type I atelocollagen and amino acid complex – structural rebuilding

Type I atelocollagen in S-Line Smooth is the same class of substance that forms the basis of the acclaimed NITHYA Type I Collagen biostimulator. Atelocollagen (without telopeptides) is minimally immunogenic — the body accepts it without an inflammatory reaction. After injection it acts as a direct substrate for fibroblasts and activates them through mechanotransduction, stimulating neocollagenogenesis.

The amino acid complex (proline, glycine, hydroxyproline, lysine and others) provides fibroblasts with direct building blocks for collagen chain synthesis, increasing the efficiency of neocollagenogenesis and accelerating skin revitalisation.

S-Line Smooth vs S-Line Shield – how do the two products differ?

• NITHYA S-Line Shield (succinic acid + HA) — acts at the level of cellular metabolism: activates the Krebs cycle, increases ATP production, provides antioxidant protection. Indicated for dull complexion, under-eye circles, cellular hypoxia.
• NITHYA S-Line Smooth (Argireline® + atelocollagen + amino acids) — acts at the neuromuscular and structural level: relaxes facial muscles (Argireline®) and rebuilds the collagen matrix. Indicated for expression wrinkles, loss of elasticity and as a botulinum toxin alternative.

Both products complement each other perfectly and can be used alternately or as a combined protocol for comprehensive biorevitalisation and rejuvenation.

FAQ – questions and answers

Is Argireline® the same as botulinum toxin (botox)?

No — the mechanism of action is similar in its goal (muscle relaxation), but completely different in the way it works. Botulinum toxin (BoNT-A) irreversibly cleaves the SNAP-25 protein, completely blocking acetylcholine release — hence the "frozen face" effect lasting 3–6 months. Argireline® works reversibly, competitively disrupting the SNARE complex — the effect is gentler, more natural and gradually fades. It is a safe, non-toxic alternative for patients who are afraid of botulinum toxin or have contraindications to it.

How does S-Line Smooth differ from Synerlift DMAE & PDRN?

NITHYA Synerlift DMAE & PDRN acts via DMAE (acetylcholine precursor, indirectly tones muscles) and PDRN (regeneration via A2A receptors) — the effect is lifting firming of the skin. S-Line Smooth acts oppositely — Argireline® relaxes muscles, reducing expression grooves. Both can be combined: Smooth for expression wrinkles, Synerlift as general lifting.

When are the results visible?

Initial smoothing of expression wrinkles is visible after 2–3 sessions. The full effect of muscle relaxation and collagen rebuilding builds throughout the full 4–6 session cycle. The effect is maintained for several weeks after the cycle is completed.

Can S-Line Smooth be used during dermapen treatment?

Yes — the product is suitable for both needle and microneedling (dermapen) mesotherapy. In microneedling it is applied as a serum on the skin during the procedure. Needle mesotherapy is however the recommended primary technique due to the higher delivery of Argireline® in proximity to neuromuscular junctions.

Can S-Line Smooth be combined with other Nithya products?

Yes — the product fits perfectly into comprehensive rejuvenation protocols of the Nithya line. Recommended combined protocols: S-Line Smooth + NITHYA Type I Collagen (Smooth relaxes muscles, collagen rebuilds the deep matrix) or S-Line Smooth + S-Line Shield (Smooth smooths expression wrinkles, Shield activates metabolism and hydrates).
