What is NITHYA S-Line Stimulate?

NITHYA S-Line Stimulate is an intensively biostimulating preparation for needle mesotherapy produced by Euroresearch Srl. It is the third pillar of the S-Line family, addressing a different mechanism of ageing than its sister preparations — S-Line Stimulate focuses on deep fibroblast stimulation and extracellular matrix remodelling via the TGF-β1 pathway. Particularly indicated for advanced ageing, loss of skin density, stretch marks and scars.

The formula combines three complementary active substances: Tetrapeptide-2 (biomimetic TGF-β1 activator), type I atelocollagen (fibroblast mechanotransduction and biostimulation) and sodium hyaluronate (hydration and ECM support).

Tetrapeptide-2 – mechanism of action via the TGF-β1 pathway

Tetrapeptide-2 is a synthetic biomimetic peptide (4 amino acid chain) that mimics the action of thymopoietin — a hormone produced by the thymus gland involved in immune regulation and skin cell differentiation. In the aesthetic medicine context, Tetrapeptide-2 is distinguished by a dual anti-ageing mechanism:

• Activation of the TGF-β1 pathway (Transforming Growth Factor β1): Tetrapeptide-2 stimulates β1-integrin receptors on fibroblasts, triggering the TGF-β1 signalling cascade — the primary regulator of collagen synthesis in the dermis. Effect: fibroblasts intensify production of type I and III collagen, elastin and fibronectin.
• MMP inhibition (Matrix Metalloproteinases): Tetrapeptide-2 reduces the activity of MMP enzymes responsible for collagen degradation. The effect is therefore bilateral — more collagen is synthesised AND less is degraded, significantly accelerating dermal matrix remodelling.
• Fibroblast proliferation stimulation — Tetrapeptide-2 promotes fibroblast cell division, increasing their number and metabolic activity. The skin gains more active collagen-producing cells.
• ECM remodelling — simultaneous synthesis activation and degradation inhibition leads to densification of the extracellular matrix, translating into measurable improvement in skin density and elasticity.

Thanks to this mechanism, Tetrapeptide-2 is particularly effective for stretch marks and scars — areas where permanent damage to the collagen structure of the ECM has occurred and active matrix remodelling is required.

Type I atelocollagen – synergy with Tetrapeptide-2

Type I atelocollagen in S-Line Stimulate acts on fibroblasts through mechanotransduction — physical contact of collagen with fibroblast integrin receptors triggers internal signalling pathways, independently of Tetrapeptide-2. Together they create dual fibroblast stimulation through two different receptor mechanisms, multiplying the biostimulatory effect. This is the same class of active substance as in the acclaimed NITHYA Type I Collagen — here enriched with Tetrapeptide-2.

Sodium hyaluronate – hydration and ECM environment

Sodium hyaluronate complements the action of Tetrapeptide-2 and atelocollagen: it hydrates the skin intradermally, provides an optimal environment for regenerative processes, improves elasticity and turgor, and reduces wrinkle depth through a hydration effect in the extracellular matrix.

FAQ – questions and answers

How does Tetrapeptide-2 differ from Argireline® (Hexapeptide-8)?

These are two completely different peptides acting on different targets. Argireline® (in S-Line Smooth) is a SNARE complex inhibitor — it relaxes facial muscles, reducing dynamic wrinkles. Tetrapeptide-2 (in S-Line Stimulate) activates the TGF-β1 pathway in fibroblasts — intensively stimulating collagen synthesis and inhibiting its degradation. Argireline® works "from above" (relaxes muscles), Tetrapeptide-2 works "from below" (rebuilds the dermal structure).

Is S-Line Stimulate effective for stretch marks?

Yes — stretch marks (striae distensae) are areas where permanent damage to the collagen structure of the dermis has occurred. Tetrapeptide-2 through TGF-β1 activation and MMP inhibition stimulates active remodelling of damaged collagen. Effects are gradual and require regular treatment cycles, but Stimulate is one of the few mesotherapy preparations that mechanistically addresses the cause of stretch marks — not just their appearance.

How does S-Line Stimulate differ from NITHYA Type I Collagen?

NITHYA Type I Collagen is pure atelocollagen — exceptionally concentrated biostimulation through mechanotransduction. S-Line Stimulate combines atelocollagen with Tetrapeptide-2 (TGF-β1) and HA — it is a broader formula combining collagen biostimulation with hydration and additional stimulation through the peptide pathway. Both preparations can be used in combined protocols.

When are the results visible?

Improved skin hydration and radiance are visible after 1–2 sessions (HA effect). Collagen biostimulation effects (densification, improved elasticity, wrinkle depth reduction) build gradually — full effect after 4–8 weeks following completion of the cycle. Reduction of stretch marks and scars requires regular series and a longer observation period.

Can S-Line Stimulate be combined with other Nithya products?

Yes.
