What is Nucleofill Soft Plus Eyes?

Nucleofill Soft Plus Eyes is a specialist product from the Promoitalia Group SpA range (Milan, Italy) based on polymerized polynucleotides (PN) at a concentration of 7.5 mg/ml. It is the only product in this line dedicated exclusively to the exceptionally sensitive skin of the periorbital area — a zone where most standard mesotherapy preparations are too viscous, too irritating or simply not anatomically adapted. The product is administered using a 30G needle directly into the subcutaneous tissue of the periorbital area.

Polynucleotides (PN) vs PDRN — how does Nucleofill differ from PDRN products?

Both PDRN (polydeoxyribonucleotide, e.g. Rejuran) and PN (polynucleotides, e.g. Nucleofill) are DNA derivatives obtained from salmon or trout roe. The difference is however clinically significant:

• PDRN — short DNA fragments with a molecular weight of 50–1500 kDa. Excellent at activating A2A receptors, acts quickly at the level of cellular signalling. High fluidity — easy to inject.
• PN (polymerized polynucleotides) — longer, polymerized DNA chains with a significantly higher molecular weight. They form a three-dimensional gel network within the tissue that simultaneously acts as a biological hydration scaffold (iso-osmotic hydration of the ECM) and as a receptor biostimulator. The hydration effect is more durable and deeper than with PDRN.

Nucleofill Soft Plus Eyes uses a polymerized form of PN — after injection into the tissue the product forms a three-dimensional polynucleotide network that not only stimulates fibroblasts but physically retains water in the extracellular matrix for significantly longer than standard PDRN.

Triple-mechanism action of Nucleofill – biostimulation, antioxidation, hydration

Promoitalia describes the mechanism of action of Nucleofill as three overlapping processes:

• Stage 1 — Biostimulation (activation of collagen and elastin synthesis): polynucleotides bind to A2A receptors on fibroblast membranes, triggering signalling pathways responsible for the synthesis of type I collagen and elastin. Effect: dermal densification, lifting, improved elasticity and skin tone around the eyes.
• Stage 2 — Antioxidation (fighting oxidative stress): PN is a potent free radical scavenger. The periorbital area is particularly exposed to oxidative stress — eyelids are constantly exposed to UV radiation, and eye movements and blinking generate continuous micro-mechanical stress in the tissue. Antioxidant protection slows the degradation of collagen and elastin fibres.
• Stage 3 — Iso-osmotic ECM hydration: the polymerized PN network forms a three-dimensional matrix retaining water in the extracellular matrix. Unlike HA, which binds water osmotically, PN hydrates iso-osmotically — in a way consistent with tissue physiology and significantly safer for the delicate periorbital skin.

Why does the eye area require a dedicated product?

The skin of the periorbital area is anatomically and physiologically completely different from the skin of the rest of the face — which is precisely why standard HA fillers and most mesotherapy preparations cannot be used here:

• Skin thickness: ~0.5 mm vs ~2 mm in other facial areas. The smallest technical error, too viscous a product or too large a volume — and the result is a visible lump, swelling or irregularity.
• Orbicularis oculi muscle — works continuously with every blink (~15,000 times a day). This constant contraction causes accelerated degradation of collagen and elastin fibres in this area.
• Poor lymphatic drainage — predisposes to fluid accumulation and the formation of under-eye puffiness. Any substance injected here must be exceptionally well absorbed and must not impede drainage.
• Dark circles — caused by: (a) blood vessels showing through thin skin, (b) haemosiderin deposits after microextravasations, (c) hypoxia and microcirculation disorders. Nucleofill directly addresses the microcirculatory and hypoxic component.

How does Nucleofill Soft Plus Eyes differ from a standard HA skinbooster for the eye area?

HA skinboosters hydrate and fill — they act solely mechanically and osmotically. Nucleofill additionally stimulates collagen biogenesis via A2A receptors, provides antioxidant protection and hydrates iso-osmotically — more safely for the thin periorbital skin. How does Nucleofill differ from PDRN products (e.g. Rejuran)?
